Sonodynamic therapy for treatment of C6 glioma in a rat model step 1: feasibility of tumor model

چکیده
Background/introduction Gliomas represent the most common and devastating primary CNS tumor. While surgical resection is the mainstay of treatment, treating physicians are seeking a safer and more effective treatment modality that is noninvasive, deep-penetrating, and less prone to infection and damage to surrounding tissues. Photodynamic therapy, utilizing a photosensitizing agent with laser light, while effective in a variety of solid tissue tumors, has not proven to be especially advantageous in the treatment of gliomas, due to the poor penetration of the light. Sonodynamic therapy has been explored as an alternative to PDT. Sensitizers such as 5-aminolevulinic acid (5-ALA) and indocyanine green (IcG) have been shown to be preferentially taken up by glioma cells. Low-intensity ultrasound waves can provide enough energy to activate sensitizing agents and induce apoptosis, without the collateral damage to surrounding tissues seen with laser light in PDT. The aim of this initial study is to determine whether the sonosensitizers 5ALA and IcG can be effectively delivered to, and detected in, the subcutaneous C6 glioma rat model.

Study of the mechanism of sonodynamic therapy in a rat glioma model
PURPOSE The study reported here examined the effect of hematoporphyrin monomethyl ether (HMME)-mediated sonodynamic therapy (SDT) on C6 gliomas implanted in rat brains. METHODS Two weeks after inoculation, glioma development was evaluated by measuring tumor volume using a 1.5 T magnetic resonance imager. Rats that had a well-developed C6 glioma (usually when the tumor diameter reached 3-5 mm)...
متن کاملEvaluation of the Combined Effects of Sonodynamic and Photodynamic Therapies in a Colon Carcinoma Tumor Model (CT26)
Introduction: Photodynamic therapy is a noninvasive therapeutic method for tumors with a maximum depth of 5 mm. On the other hand, most photosensitizers are also susceptible to ultrasound waves (the basis of sonodynamic therapy). Therefore, it is expected that a combination of the two therapeutic methods will increase effectiveness of photodynamic therapies for lower doses of sensitizer and cur...
متن کاملEffects of Combined Sonodynamic and Photodynamic Therapies on a Colon Carcinoma Tumor Model
Objective(s) Although photodynamic therapy is considered as a noninvasive method, most photosensitizers are susceptible to ultrasound. Therefore, it is expected that the combination of two activation methods might have a synergistic effect. This probable effect has been investigated in this study. Materials and Methods This study was conducted on colon carcinoma ...
متن کاملLow Frequency Ultrasonication Induced Antitumor Effect in 5-Aminolevulinic Acid Treated Malignant Glioma
We investigated the feasibility of sonodynamic therapy for glioma by low frequency ultrasound with 5-aminolevulinic acid (5-ALA), a precursor of protoporphyrin IX (PpIX) in heme synthetic process. In vivo tumor model was made by inoculating human glioma cell line U87-MG subcutaneously in nude mice. The tumor was sonicated by 25-kHz ultrasound 4 hours following administration of 5-ALA. The tumor...
متن کاملSonophotodynamic therapy mediated by liposomal zinc phthalocyanine in a colon carcinoma tumor model: Role of irradiating arrangement
Objective(s): Low penetration depth of light is the main defect of photodynamic therapy (PDT), which could be improved by sonodynamic therapy (SDT). In this study, a combination of PDT and SDT known as sonophotodynamic therapy (SPDT) was investigated using two reverse arrangements in CT26 tumor model. Materials and Methods: The liposomal zinc phthalocyanine was synthesized and characterized. It...
